The ACTN3 gene encodes alpha-actinin-3. a sarcomeric protein predominantly expressed in fast-twitch skeletal muscle fibers, where it stabilizes the contractile apparatus and regulates energy metabolism. A common polymorphism (R577X) results in a premature stop codon, causing ACTN3 deficiency in approximately 20% of the global population. This genetic variation has been linked to athletic performance, with the functional allele (577R) associated with power/sprint-oriented traits, while the null variant (577X) correlates with endurance adaptations.

ACTN3 antibodies are essential tools for studying protein expression, localization, and function in muscle biology. They enable researchers to investigate ACTN3's role in muscle physiology, its interaction with dystrophin-associated proteins, and its implications in conditions like muscular dystrophy, sarcopenia, and metabolic disorders. Commercial ACTN3 antibodies are typically developed using immunogenic peptide sequences unique to the ACTN3 isoform, ensuring specificity over the homologous ACTN2 protein.

Applications include Western blotting, immunohistochemistry, and immunofluorescence to assess ACTN3 expression levels in tissue samples, cell cultures, or genetically modified models.
