The SLC4A4 gene encodes the sodium bicarbonate cotransporter NBCe1. a critical protein involved in maintaining pH homeostasis and ion transport across cell membranes. Expressed predominantly in the kidneys, pancreas, and eyes, NBCe1 facilitates the coupled movement of sodium and bicarbonate ions, playing essential roles in acid-base balance, pancreatic bicarbonate secretion, and aqueous humor production. Dysfunctional NBCe1 is linked to diseases such as proximal renal tubular acidosis (pRTA), glaucoma, and diabetes.

SLC4A4 antibodies are tools used to study the expression, localization, and function of NBCe1 in both physiological and pathological contexts. These antibodies are employed in techniques like Western blotting, immunohistochemistry, and immunofluorescence to detect NBCe1 isoforms (NBCe1-A, -B, -C) in tissue-specific studies. Research utilizing SLC4A4 antibodies has elucidated mechanisms underlying NBCe1-related disorders, such as impaired bicarbonate reabsorption in pRTA or dysregulated intraocular pressure in glaucoma.

Additionally, SLC4A4 antibodies aid in diagnosing genetic mutations affecting NBCe1. particularly in cases of inherited pRTA with ocular abnormalities. Their application extends to exploring therapeutic targets, such as modulating transporter activity in metabolic diseases. However, challenges remain in ensuring antibody specificity due to structural similarities among SLC4 family members. Continued development and validation of SLC4A4 antibodies are vital for advancing understanding of bicarbonate transport biology and associated diseases.
