The ribosomal protein lateral stalk subunit P0 (RPLP0), also known as acidic ribosomal phosphoprotein P0 or 36B4. is a key component of the 60S ribosomal subunit. It plays a structural and functional role in translation by forming the ribosomal "stalk" structure, which facilitates GTPase-dependent interactions during protein synthesis. RPLP0 is highly conserved across eukaryotes and is constitutively expressed in most tissues, making it a common reference ("housekeeping") gene in gene expression studies.

Antibodies targeting RPLP0 are widely used in research to study ribosome biogenesis, translational regulation, and cellular stress responses. They serve as critical tools for Western blotting, immunofluorescence, and immunohistochemistry to validate protein loading controls or investigate ribosomal dysfunction in diseases like cancer, neurodegenerative disorders, and ribosomopathies. However, its role as a loading control has been debated due to variable expression under certain conditions, such as cellular differentiation or drug treatment. Recent studies also implicate RPLP0 in extraribosomal functions, including cell cycle regulation and apoptosis. Commercial RPLP0 antibodies are typically validated against conserved epitopes, though species-specific cross-reactivity should be confirmed experimentally.
