Neurotrophin-4 (NTF4), also known as NT-4 or NT-5 depending on species, is a member of the neurotrophin family of growth factors that play critical roles in neuronal survival, development, and plasticity. It binds to the tropomyosin receptor kinase B (TrkB) and the low-affinity p75 neurotrophin receptor, activating downstream signaling pathways involved in synaptic modulation, axon guidance, and cell differentiation. NTF4 is expressed in both the central and peripheral nervous systems and has been implicated in processes such as memory formation, pain regulation, and recovery from neural injury. Dysregulation of NTF4 signaling has been linked to neurodegenerative diseases (e.g., Alzheimer’s), mood disorders, and neuropathic pain.

Antibodies targeting NTF4 are essential tools for studying its expression, localization, and function in biological systems. They are widely used in techniques like Western blotting, immunohistochemistry, and ELISA to quantify protein levels or visualize distribution in tissues. Commercial NTF4 antibodies are typically raised in hosts such as rabbits or mice using recombinant protein fragments or synthetic peptides. However, challenges remain in ensuring specificity due to high homology among neurotrophin family members. Recent research emphasizes the need for rigorous validation via knockout controls or neutralization assays. Such antibodies have advanced understanding of NTF4's role in neuroprotection and its potential as a therapeutic target, particularly in conditions involving neuronal degeneration or synaptic dysfunction.
