The MASP1 (Mannan-Binding Lectin Serine Protease 1) antibody targets a key enzyme in the lectin pathway of the complement system. MASP1 is synthesized primarily in the liver and circulates as a zymogen bound to pattern-recognition molecules like MBL (mannose-binding lectin), ficolins, and collectins. Upon pathogen-associated molecular pattern recognition, MASP1 undergoes autoactivation, playing a dual role: it cleaves complement components C4 and C2 to initiate the complement cascade, and exhibits thrombin-like activity, contributing to coagulation and inflammatory responses.

MASP1 antibodies are essential tools for studying its expression, activation mechanisms, and interactions in immune regulation. Dysregulation of MASP1 is linked to diseases, including 3MC syndrome (a developmental disorder caused by genetic mutations) and inflammatory or autoimmune conditions. Research applications include immunohistochemistry, Western blotting, and functional assays to explore its role in host defense, immune-complex disorders, and potential therapeutic targeting. Commercial antibodies are typically raised against specific epitopes of human MASP1. enabling isoform-specific detection given its structural homology with MASP2 and MAp44. Recent studies also investigate its non-complement functions in embryogenesis and tissue remodeling.
