LYVE-1 (Lymphatic Vessel Endothelial Hyaluronan Receptor 1) is a transmembrane glycoprotein primarily expressed on lymphatic endothelial cells, serving as a key marker for distinguishing lymphatic vessels from blood vessels. It belongs to the CD44 hyaluronan receptor family and binds hyaluronan, a glycosaminoglycan involved in cell adhesion, migration, and tissue remodeling. Discovered in the late 1990s, LYVE-1 plays a role in lymphatic development, immune cell trafficking, and tumor metastasis via the lymphatic system. Antibodies targeting LYVE-1 are widely used in research to visualize lymphatic networks in tissues, study lymphatic-related diseases (e.g., lymphedema, cancer metastasis), and explore immune responses. These antibodies are critical tools in immunohistochemistry, flow cytometry, and immunofluorescence, helping differentiate lymphatic structures from vascular ones (e.g., blood vessels marked by CD31 or Podoplanin). However, LYVE-1 expression isn’t exclusive to lymphatics; it’s also found in liver sinusoidal endothelial cells and activated macrophages, necessitating careful experimental validation. Its conserved structure across species supports translational studies in murine models. Ongoing research focuses on LYVE-1’s dual roles in hyaluronan metabolism and immune regulation, highlighting its therapeutic potential in lymphatic disorders and cancer.
