Histone deacetylase 8 (HDAC8) is a class I zinc-dependent deacetylase involved in epigenetic regulation by removing acetyl groups from lysine residues on histones and non-histone proteins. It localizes to both the nucleus and cytoplasm, distinguishing it from other class I HDACs (e.g., HDAC1-3) that are primarily nuclear. HDAC8 plays critical roles in cell cycle progression, transcriptional regulation, and chromatin remodeling, with emerging links to developmental processes, metabolic pathways, and immune responses. Dysregulation of HDAC8 is implicated in cancers (e.g., neuroblastoma, leukemia), Cornelia de Lange syndrome (CdLS, linked to mutations in HDAC8), and neurological disorders.

HDAC8-specific antibodies are essential tools for studying its expression, localization, and functional interactions. These antibodies enable detection via techniques like Western blotting, immunohistochemistry (IHC), immunofluorescence (IF), and chromatin immunoprecipitation (ChIP). Due to structural similarities among HDAC isoforms, antibody specificity is critical to avoid cross-reactivity. Validated HDAC8 antibodies help investigate its role as a therapeutic target, particularly in evaluating HDAC inhibitors (e.g., selective HDAC8 inhibitors in preclinical cancer studies). Recent research also explores HDAC8's non-canonical substrates, expanding its biological relevance beyond epigenetic regulation.
