The TAP2 (Transporter Associated with Antigen Processing 2) protein is a critical component of the major histocompatibility complex (MHC) class I antigen presentation pathway. It forms a heterodimeric complex with TAP1. functioning as an ATP-binding cassette (ABC) transporter located in the endoplasmic reticulum (ER) membrane. Together, TAP1 and TAP2 mediate the translocation of cytosolic antigenic peptides into the ER lumen, where they bind to MHC class I molecules for subsequent presentation to CD8+ T cells, enabling immune surveillance of infected or cancerous cells.

Antibodies targeting TAP2 are essential tools for studying its expression, localization, and functional interactions. They are widely used in techniques like Western blotting, immunofluorescence, and flow cytometry to investigate TAP2 regulation under pathological conditions, such as viral immune evasion (e.g., herpesviruses downregulate TAP2 to inhibit antigen presentation) or autoimmune disorders. Mutations in the TAP2 gene are linked to bare lymphocyte syndrome type I (MHC class I deficiency), characterized by recurrent infections. Research using TAP2 antibodies has also explored its role in cancer immunology, as dysregulated antigen presentation may contribute to tumor immune escape. These studies underscore TAP2's importance in adaptive immunity and its potential as a therapeutic or diagnostic target.
