NLRP12 (NACHT, LRR, and PYD domains-containing protein 12) is a member of the nucleotide-binding oligomerization domain (NOD)-like receptor (NLR) family, which plays a role in innate immune responses. Primarily expressed in myeloid cells, NLRP12 functions as a regulatory protein in inflammatory signaling pathways. It modulates NF-κB and MAPK activation, often acting as a negative regulator to suppress excessive inflammation. NLRP12 also participates in inflammasome formation under specific conditions, though its role here is less characterized compared to other NLRs like NLRP3.

Mutations in the NLRP12 gene have been linked to autoinflammatory disorders, such as familial cold autoinflammatory syndrome-2 (FCAS2), highlighting its importance in immune homeostasis. NLRP12 antibodies are critical tools for studying its expression, localization, and interactions in both physiological and pathological contexts. These antibodies (monoclonal or polyclonal) are widely used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to investigate NLRP12's involvement in inflammatory diseases, cancer, and microbial infections. Recent studies also explore its dual role in promoting or suppressing inflammation depending on cellular context, making NLRP12 a compelling target for therapeutic research.
