The histamine H3 receptor (HRH3) is a G protein-coupled receptor predominantly expressed in the central nervous system, where it regulates neurotransmitter release via presynaptic autoreceptors and heteroreceptors. Discovered in 1983. HRH3 plays a key role in modulating cognitive functions, sleep-wake cycles, and energy homeostasis. HRH3 antibodies are essential tools for studying receptor localization, expression levels, and signaling mechanisms in both physiological and pathological contexts. These antibodies enable detection of HRH3 in tissues or cells through techniques like immunohistochemistry, Western blotting, and flow cytometry. Research using HRH3 antibodies has linked receptor dysfunction to neurological disorders such as narcolepsy, Alzheimer’s disease, and attention deficit hyperactivity disorder (ADHD). Additionally, HRH3 is explored as a therapeutic target, with antagonists investigated for cognitive enhancement and agonists for sleep regulation. Validating antibody specificity remains critical due to structural similarities among histamine receptor subtypes.
