Natural Cytotoxicity Triggering Receptor 2 (NCR2), also known as NKp44. is a cell surface receptor predominantly expressed on natural killer (NK) cells, a key component of the innate immune system. Encoded by the *NCR2* gene, this activating receptor plays a critical role in NK cell-mediated cytotoxicity against virus-infected and malignant cells. Structurally, NCR2 belongs to the immunoglobulin superfamily, featuring a single extracellular V-type domain, a transmembrane region, and a short cytoplasmic tail that associates with adaptor proteins like DAP12 to transmit activation signals. Unlike other NCR family members (NKp30. NKp46), NKp44 is expressed only upon NK cell activation, making it a marker for functional NK subsets.

NCR2 antibodies are tools used to study receptor function or modulate NK cell activity. Research highlights its interaction with ligands such as proliferating cell nuclear antigen (PCAN) on stressed cells, viral proteins, and components of the tumor microenvironment. Dysregulated NCR2 expression is linked to impaired immune surveillance in cancers, infections, and autoimmune diseases. Therapeutic NCR2 antibodies are being explored to enhance NK cell anti-tumor responses or block pathogenic signaling. Additionally, soluble NCR2 variants and genetic polymorphisms are studied for diagnostic or prognostic value in diseases like leukemia or COVID-19. Overall, NCR2 antibodies serve as pivotal reagents in unraveling NK cell biology and advancing immunotherapeutic strategies.
