The ZAP70 (zeta-chain-associated protein kinase 70) antibody is a critical tool for studying the role of ZAP70. a protein tyrosine kinase essential in T-cell receptor (TCR) signaling. ZAP70 is part of the Syk family of kinases and is primarily expressed in T cells and natural killer (NK) cells. Upon TCR activation, ZAP70 is recruited to phosphorylated immunoreceptor tyrosine-based activation motifs (ITAMs) on the CD3ζ chain, leading to its activation. This triggers downstream signaling cascades, including calcium flux, MAPK pathways, and NF-κB activation, which are vital for T-cell differentiation, proliferation, and immune response.

Mutations in the ZAP70 gene are linked to severe combined immunodeficiency (SCID), characterized by dysfunctional T cells and recurrent infections. In research, ZAP70 antibodies are used to detect protein expression via techniques like Western blot, flow cytometry, and immunohistochemistry. Clinically, ZAP70 expression serves as a prognostic marker in chronic lymphocytic leukemia (CLL), where its presence correlates with aggressive disease and poorer outcomes.
