Lumican (LUM), a member of the small leucine-rich proteoglycan (SLRP) family, is a key extracellular matrix protein predominantly expressed in connective tissues such as the cornea, skin, and cartilage. It plays a critical role in regulating collagen fibril organization, maintaining tissue transparency (e.g., in the cornea), and modulating cell adhesion, migration, and signaling. Dysregulation of lumican has been implicated in pathologies including corneal dystrophy, fibrosis, and cancer, where it exhibits dual roles as either a tumor suppressor or promoter depending on context.

LUM antibodies are immunological tools developed to detect and quantify lumican in research and diagnostic applications. These antibodies are typically generated using recombinant lumican proteins or peptide epitopes as immunogens in animal hosts. They enable the study of lumican's spatial distribution (via immunohistochemistry), expression levels (via Western blot or ELISA), and interactions in tissue homeostasis and disease. Commercial LUM antibodies are often validated for cross-reactivity across species (human, mouse, rat) and tested for specificity to avoid cross-reactivity with other SLRPs like decorin or fibromodulin. Recent studies utilizing LUM antibodies have highlighted its potential as a biomarker in cancer prognosis and stromal remodeling, underscoring its therapeutic and diagnostic relevance.
