UBE2S (Ubiquitin-Conjugating Enzyme E2S) is a member of the ubiquitin-conjugating enzyme family, which plays a critical role in the ubiquitin-proteasome system (UPS) responsible for targeted protein degradation. As an E2 enzyme, UBE2S facilitates the transfer of ubiquitin molecules to specific substrate proteins, typically in coordination with E3 ubiquitin ligases. This process, known as ubiquitination, tags proteins for proteasomal destruction or modulates their cellular localization and activity. UBE2S is particularly notable for its role in catalyzing the formation of K11-linked polyubiquitin chains, a specialized modification associated with cell cycle regulation and mitotic progression. Studies highlight its interaction with the anaphase-promoting complex/cyclosome (APC/C), where it contributes to the degradation of mitotic regulators, ensuring proper chromosome segregation and cell division.

In cancer research, UBE2S has garnered attention due to its frequent overexpression in various malignancies, including breast, liver, and colorectal cancers. Elevated UBE2S levels correlate with tumor aggressiveness, therapeutic resistance, and poor prognosis, suggesting its potential as a biomarker or therapeutic target.
