MPG (N-Methylpurine-DNA Glycosylase), also known as AAG (alkyladenine DNA glycosylase), is a DNA repair enzyme critical for maintaining genomic stability. It initiates the base excision repair (BER) pathway by recognizing and excising damaged or aberrant DNA bases, including alkylated (e.g., 3-methyladenine, 7-methylguanine) and oxidized purines. This repair mechanism is essential for counteracting mutagenic and cytotoxic effects caused by endogenous metabolic byproducts, environmental toxins, or chemotherapeutic agents. MPG's role in DNA repair has linked it to cancer biology, as dysregulation or mutations in MPG may influence cancer susceptibility, therapeutic resistance, or response to DNA-damaging treatments.

MPG antibodies are tools developed to study the expression, localization, and function of this enzyme in biological systems. They are widely used in techniques such as Western blotting, immunohistochemistry, and immunofluorescence to quantify MPG levels in tissues or cell lines, particularly in cancer research. Studies have explored MPG's dual role—while its repair activity protects against carcinogenesis, overexpression in tumors may enhance repair of chemotherapy-induced DNA damage, contributing to drug resistance. Additionally, MPG polymorphisms are investigated for associations with disease risk. Research utilizing MPG antibodies continues to clarify its therapeutic potential, either as a biomarker for treatment stratification or a target to sensitize cancer cells to genotoxic therapies.
