The EIF2AK4 antibody is a tool used to detect eukaryotic translation initiation factor 2 alpha kinase 4 (EIF2AK4), also known as GCN2. a serine/threonine kinase critical in cellular stress responses. EIF2AK4 regulates the integrated stress response (ISR) by phosphorylating the alpha subunit of eukaryotic initiation factor 2 (eIF2α) under conditions such as amino acid deprivation, UV exposure, or viral infection. This phosphorylation halts global protein synthesis while promoting the translation of stress-responsive genes, enabling cells to adapt to adverse conditions.

Research on EIF2AK4 has gained attention due to its role in diseases like pulmonary arterial hypertension (PAH). Biallelic mutations in the EIF2AK4 gene are linked to heritable pulmonary veno-occlusive disease/pulmonary capillary hemangiomatosis (PVOD/PCH), a rare and severe form of PAH. Antibodies targeting EIF2AK4 are essential for studying its expression, activation, and interaction networks in both physiological and pathological contexts. They are widely used in techniques like Western blotting, immunohistochemistry, and immunofluorescence to explore its tissue distribution, signaling pathways, and potential therapeutic targeting.

Understanding EIF2AK4’s function through antibody-based assays may unveil novel insights into stress adaptation mechanisms and inform treatments for conditions involving dysregulated ISR, including cancer, neurodegenerative disorders, and metabolic diseases.
