NEK2 (NIMA-related kinase 2) is a serine/threonine kinase belonging to the NEK family, which plays a critical role in regulating cell cycle progression, particularly during the G2/M phase transition and mitosis. It is involved in centrosome maturation, spindle assembly, and chromosome alignment, ensuring genomic stability. Dysregulation of NEK2 is linked to various cancers, where its overexpression correlates with tumor progression, chemotherapy resistance, and poor prognosis. NEK2 activity is tightly controlled through phosphorylation and ubiquitination, with its protein levels peaking during mitosis before degradation via the ubiquitin-proteasome system.

NEK2 antibodies are essential tools for studying its expression, localization, and function in cellular processes. They are widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry. Due to the presence of NEK2 isoforms (e.g., NEK2A and NEK2B), antibodies are often designed to target specific regions, such as the kinase domain or unique C-terminal sequences, to ensure isoform specificity. Some antibodies also detect phosphorylation states (e.g., Thr175), aiding in the study of NEK2 activation mechanisms.
