The P2RY12 antibody is a research tool targeting the P2Y purinoceptor 12. a G protein-coupled receptor predominantly expressed on microglia in the central nervous system. P2RY12 plays a critical role in microglial chemotaxis, surveillance, and response to extracellular nucleotides like ADP released during cellular damage. As a Gi-coupled receptor, it inhibits cAMP signaling and modulates ion channel activity, influencing microglial migration toward injury sites. Antibodies against P2RY12 are widely used to identify and study microglial populations, particularly in distinguishing homeostatic ("resting") microglia from activated states in neuroinflammatory or degenerative conditions.

Recent studies highlight P2RY12's involvement in neurodegenerative diseases, including Alzheimer's disease and multiple sclerosis, where microglial dysfunction contributes to pathology. The antibody aids in mapping microglial heterogeneity and activation patterns in human postmortem tissues and animal models. Interestingly, decreased P2RY12 expression correlates with chronic neuroinflammation, making it a potential biomarker for disease progression. Therapeutic strategies exploring P2RY12 antagonists (e.g., antithrombotic drugs like clopidogrel) have expanded into neurological research, investigating dual benefits in vascular and neuroimmune modulation. However, the receptor's dual roles in neuroprotection and potential neurotoxicity remain under investigation, emphasizing the need for precise tools like P2RY12 antibodies to unravel context-dependent microglial behaviors.
