ELANE (Elastase, Neutrophil Expressed) encodes neutrophil elastase, a serine protease primarily secreted by neutrophils during inflammation. This enzyme plays a critical role in innate immunity by degrading pathogens and remodeling extracellular matrix components. Dysregulation of ELANE is linked to several disorders. Mutations in the ELANE gene are a major cause of severe congenital neutropenia (SCN) and cyclic neutropenia, characterized by recurrent infections due to insufficient mature neutrophils. Additionally, excessive neutrophil elastase activity is implicated in chronic inflammatory conditions like chronic obstructive pulmonary disease (COPD), acute respiratory distress syndrome (ARDS), and cystic fibrosis, where tissue damage results from uncontrolled proteolytic activity.

ELANE antibodies are essential tools in research and diagnostics. They enable detection of neutrophil elastase expression in immune cells or tissues via techniques like Western blot, immunohistochemistry, and flow cytometry. These antibodies help study disease mechanisms, monitor protein levels in clinical samples, or evaluate therapeutic interventions targeting neutrophil elastase. Recent therapeutic strategies also explore inhibitory antibodies to neutralize ELANE activity, aiming to mitigate protease-driven tissue injury in inflammatory diseases. Understanding ELANE's dual role—both in host defense and pathology—highlights its significance as a biomarker and therapeutic target.
