The MUC5AC antibody is a crucial tool in studying mucin dynamics, particularly targeting the MUC5AC glycoprotein, a major component of mucosal secretions. MUC5AC, encoded by the *MUC5AC* gene, is a gel-forming mucin predominantly expressed by goblet cells and submucosal glands in epithelial tissues of the respiratory, gastrointestinal, and reproductive tracts. It plays vital roles in mucosal protection, lubrication, and pathogen trapping by forming viscoelastic mucus layers. Dysregulated MUC5AC expression is implicated in pathologies like asthma, chronic obstructive pulmonary disease (COPD), cystic fibrosis, gastric disorders, and cancers (e.g., lung, pancreatic).

MUC5AC antibodies, typically monoclonal or polyclonal, are used in immunohistochemistry (IHC), Western blotting, and ELISA to localize and quantify MUC5AC in tissue samples or biological fluids. These antibodies recognize specific epitopes on the protein’s central tandem repeat or glycosylated regions, though glycosylation variability can affect detection. Research using MUC5AC antibodies has revealed its overexpression in hypersecretory conditions (e.g., allergic inflammation, viral infections) and its potential as a biomarker for disease progression or therapeutic targeting. Challenges include epitope masking due to post-translational modifications and cross-reactivity with related mucins. Standardization of antibody validation remains critical for reproducible research outcomes.
