The DNA nucleotidylexotransferase (DNTT), also known as terminal deoxynucleotidyl transferase (TdT), is a specialized DNA polymerase that catalyzes the template-independent addition of nucleotides during V(D)J recombination, a critical process for generating diverse antigen receptors in developing B and T lymphocytes. Expressed predominantly in immature lymphoid precursors, DNTT plays a vital role in adaptive immunity by enhancing antibody and T-cell receptor diversity.

DNTT antibodies are immunodetection tools targeting this enzyme, widely utilized in research and diagnostics. In clinical pathology, DNTT antibodies serve as key markers for identifying precursor lymphoid neoplasms, such as acute lymphoblastic leukemias (ALL) and lymphoblastic lymphomas, as DNTT is aberrantly expressed in malignant lymphoid blasts but absent in mature lymphocytes. Its presence helps differentiate between immature and mature lymphoid malignancies, guiding treatment strategies.

In research, DNTT antibodies are employed to study lymphocyte development, recombination mechanisms, and immune repertoire formation. They are also used in flow cytometry, immunohistochemistry, and Western blotting to characterize hematopoietic progenitors in both normal and diseased states. Additionally, DNTT expression is explored in regenerative medicine for evaluating engineered lymphoid tissues. Despite its specificity, DNTT detection requires careful interpretation, as low-level expression may occur in non-lymphoid tissues under certain conditions.
