The peroxisome proliferator-activated receptor alpha (PPARA) is a nuclear receptor transcription factor that plays a central role in regulating lipid metabolism, fatty acid β-oxidation, and glucose homeostasis. It is highly expressed in tissues with high energy demands, such as the liver, heart, and skeletal muscle. PPARA activates target genes by binding to specific DNA response elements as a heterodimer with the retinoid X receptor (RXR), responding to endogenous ligands like fatty acids and synthetic agonists such as fibrates. Its involvement in metabolic pathways makes it a key focus in studying obesity, diabetes, atherosclerosis, and non-alcoholic fatty liver disease (NAFLD).
