The Absent in Melanoma 2 (AIM2) antibody targets AIM2. a cytosolic innate immune sensor critical for detecting double-stranded DNA (dsDNA) from pathogens or damaged host cells. AIM2 belongs to the PYHIN protein family and contains a N-terminal pyrin domain (PYD) and a C-terminal HIN200 domain, enabling dsDNA binding. Upon activation, AIM2 recruits ASC (apoptosis-associated speck-like protein) and procaspase-1 to form the AIM2 inflammasome, triggering caspase-1-mediated cleavage of pro-inflammatory cytokines IL-1β and IL-18. and inducing pyroptosis. This mechanism is vital for antimicrobial defense and inflammatory regulation.

AIM2 dysregulation is linked to autoimmune diseases (e.g., lupus, psoriasis), cancers, and infections. In cancer, AIM2 may act as a tumor suppressor by promoting apoptosis or exhibit oncogenic effects depending on context. Antibodies against AIM2 are essential tools for studying its expression, localization, and function in immune responses and disease pathogenesis. They enable techniques like Western blotting, immunofluorescence, and immunohistochemistry to explore AIM2's role in inflammasome activation, DNA damage responses, and therapeutic targeting.
