The XRCC6 antibody targets the X-ray repair cross-complementing protein 6 (XRCC6), also known as Ku70. a critical component of the Ku heterodimer formed with XRCC5 (Ku80). This complex plays a central role in DNA double-strand break (DSB) repair via the non-homologous end joining (NHEJ) pathway, essential for maintaining genomic stability. Ku70/80 binds to broken DNA ends, recruiting downstream repair factors like DNA-PKcs and facilitating ligation. Beyond repair, XRCC6 participates in telomere maintenance, transcriptional regulation, and V(D)J recombination during immune diversification. Antibodies against XRCC6 are widely used in research to study DNA damage response, cancer biology (e.g., chemoresistance mechanisms), and aging-related disorders linked to genomic instability. They are also employed in autoimmune disease diagnostics, as autoantibodies against Ku proteins are biomarkers in conditions like systemic lupus erythematosus. XRCC6 antibodies, often raised in rabbits or mice, are validated for applications such as Western blotting, immunofluorescence, and chromatin immunoprecipitation (ChIP). Specificity is confirmed using knockout controls, ensuring accurate detection of the ~70 kDa protein.
