CD24. a small heavily glycosylated cell surface protein anchored via a GPI linkage, functions as a modulator of cell-cell interactions and signaling. Initially identified as a B-cell differentiation antigen, CD24 is broadly expressed in immune cells, epithelial cells, and stem cells, participating in processes like cell adhesion, proliferation, and immune regulation. Its interaction with Siglec-10 (in humans) or Siglec-G (in mice) delivers inhibitory signals that suppress immune responses, positioning CD24 as a "don't eat me" signal that helps tumors evade phagocytosis by macrophages.

CD24 antibodies, such as clones ML5 or SN3. are widely used in research and diagnostics. In oncology, CD24 overexpression correlates with poor prognosis in cancers like breast, ovarian, and hepatocellular carcinoma, making it a potential therapeutic target. Antibodies targeting CD24 are explored for blocking immune evasion or as antibody-drug conjugates (ADCs). In immunology, CD24 antibodies help dissect its role in autoimmune diseases (e.g., SLE, multiple sclerosis) by regulating T-cell activation and cytokine production.

Recent interest focuses on CD24's role in cancer stem cells and immunotherapy resistance. Challenges include its heterogeneous glycosylation patterns affecting antibody specificity and therapeutic efficacy. Ongoing studies aim to optimize CD24-targeted therapies and combinatorial approaches with existing immune checkpoint inhibitors.
