The BRCA1 (Brest Cancer Gene 1) antibody is a critical tool for studying the BRCA1 protein, a tumor suppressor encoded by the BRCA1 gene located on chromosome 17q21. Germline mutations in BRCA1 are strongly associated with hereditary breast and ovarian cancers, as the protein plays essential roles in DNA repair, genomic stability, and cell cycle regulation. BRCA1 participates in homologous recombination repair (HRR) of double-strand DNA breaks through interactions with partners like RAD51 and BARD1. forming the BRCA1-A complex.

BRCA1 antibodies are widely used in research to detect protein expression, localization, and post-translational modifications (e.g., phosphorylation) via techniques like Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF). These antibodies often target specific domains, such as the N-terminal RING domain (critical for E3 ubiquitin ligase activity) or the C-terminal BRCT domains (involved in phosphoprotein binding). Commercial antibodies vary in specificity, with common clones (e.g., MS110. D-9) validated for distinct applications.

Challenges include cross-reactivity with homologous proteins or isoforms, necessitating rigorous validation using BRCA1-deficient cell lines or knockout controls. Research applications extend to investigating BRCA1 dysfunction in cancer progression, therapy resistance (e.g., PARP inhibitor sensitivity), and biomarker studies. Recent advances also explore its role beyond DNA repair, including transcriptional regulation and chromatin remodeling.
