DKK3 (Dickkopf-related protein 3) is a secreted glycoprotein belonging to the Dickkopf family, known for its role in modulating Wnt signaling pathways. Unlike DKK1 and DKK2. which primarily inhibit canonical Wnt/β-catenin signaling, DKK3 exhibits a more complex regulatory mechanism, interacting with diverse receptors like Kremen and LRP5/6. though its precise molecular targets remain under investigation. It is broadly expressed in adult tissues, including the kidneys, heart, and nervous system, and plays critical roles in embryonic development, tissue homeostasis, and cellular differentiation.

Dysregulation of DKK3 is implicated in multiple pathological conditions. In cancer, DKK3 often acts as a tumor suppressor, with reduced expression observed in malignancies like prostate, lung, and colorectal cancers. Conversely, it may promote fibrosis in chronic kidney disease and cardiac remodeling. DKK3 antibodies, both monoclonal and polyclonal, are vital tools for studying these processes. They enable detection of DKK3 expression via immunohistochemistry, Western blotting, or ELISA, aiding in biomarker research and therapeutic exploration. Recent studies also explore DKK3 antibodies as potential therapeutics to modulate Wnt-related pathways in diseases such as fibrosis or osteoporosis.

Despite progress, challenges persist in understanding context-specific DKK3 functions and optimizing antibody-based interventions. Ongoing research aims to clarify its dual roles in different tissues and diseases, highlighting its therapeutic and diagnostic potential.
