The RAF1 antibody targets the RAF1 protein (also known as c-Raf or CRAF), a serine/threonine kinase central to the RAS-RAF-MEK-ERK/MAPK signaling pathway. This evolutionarily conserved pathway regulates critical cellular processes, including proliferation, differentiation, survival, and apoptosis. RAF1. one of three RAF kinase family members (ARAF, BRAF, RAF1), acts downstream of RAS proteins. Upon activation by RAS GTPases, RAF1 phosphorylates MEK1/2. triggering a kinase cascade that ultimately modulates gene expression. Dysregulation of RAF1 is implicated in cancers (e.g., lung, melanoma) and developmental disorders like Noonan syndrome, often due to mutations or overexpression.

RAF1-specific antibodies are essential tools for studying its expression, post-translational modifications (e.g., phosphorylation at S338/Y341), and interactions in normal and diseased states. These antibodies enable techniques such as Western blotting, immunohistochemistry, immunoprecipitation, and immunofluorescence. Researchers use them to investigate RAF1's role in oncogenic signaling, resistance to targeted therapies (e.g., BRAF inhibitors), and feedback mechanisms within the MAPK pathway. Both monoclonal and polyclonal RAF1 antibodies are available, with validation across species and applications being critical for specificity, as cross-reactivity with BRAF or ARAF can occur. Their development supports drug discovery efforts aiming to inhibit aberrant RAF1 activity in precision oncology.
