NEDD8 (Neural precursor cell-expressed developmentally downregulated 8) is a ubiquitin-like protein that plays a critical role in post-translational protein modification through a process called neddylation. This covalent conjugation of NEDD8 to target proteins, primarily cullin family members, regulates the activity of Cullin-RING E3 ubiquitin ligases (CRLs), which are essential for ubiquitination and subsequent proteasomal degradation of substrate proteins. NEDD8 shares ~60% sequence identity with ubiquitin but has distinct biochemical pathways mediated by specific E1 (activating), E2 (conjugating), and E3 (ligating) enzymes.

NEDD8 antibodies are vital tools for studying neddylation dynamics in cellular processes such as cell cycle progression, DNA repair, and stress responses. Dysregulation of neddylation is implicated in cancer, neurodegenerative diseases, and viral infections, driving interest in therapeutic targeting. Researchers use NEDD8-specific antibodies to detect neddylated proteins via techniques like Western blotting, immunofluorescence, and immunoprecipitation. These antibodies must exhibit high specificity to differentiate NEDD8 from ubiquitin and other ubiquitin-like modifiers, given their structural similarities. Recent applications include investigating MLN4924 (a NEDD8-activating enzyme inhibitor) in preclinical cancer studies and exploring neddylation's role in protein homeostasis. Validation of antibody specificity using knockdown/knockout controls or recombinant proteins remains crucial for experimental accuracy.
