The cyclin-dependent kinase 5 (CDK5) antibody is a crucial tool for studying the role of CDK5. a serine/threonine kinase belonging to the CDK family. Unlike other CDKs involved in cell cycle regulation, CDK5 is primarily active in postmitotic neurons, where it regulates neuronal development, synaptic plasticity, and neurotransmission through interactions with its neuron-specific activators, p35 and p39. Dysregulation of CDK5 activity, often linked to proteolytic conversion of p35 to p25. has been implicated in neurodegenerative diseases (e.g., Alzheimer’s and Parkinson’s), neurodevelopmental disorders, and cancer.

CDK5 antibodies are designed to detect and quantify CDK5 expression, localization, and activation states in tissues or cell lysates. They are widely used in techniques like Western blotting, immunohistochemistry, and immunoprecipitation to investigate CDK5’s physiological and pathological roles. Challenges in antibody development include ensuring specificity due to structural homology among CDK family members and distinguishing between active/inactive forms or splice variants. Validation often involves knockout controls or phosphorylation-specific epitope verification.

Commercial CDK5 antibodies are typically monoclonal or polyclonal, with variations targeting distinct epitopes (N-terminal, C-terminal, or phosphorylated residues). Researchers must select antibodies validated for their specific applications and species reactivity. Recent studies also utilize CDK5 inhibitors alongside antibodies to explore therapeutic targeting in disease models.
