The PRKDC antibody targets the protein encoded by the PRKDC gene, also known as DNA-dependent protein kinase catalytic subunit (DNA-PKcs). This large serine/threonine kinase is a critical component of the DNA-dependent protein kinase (DNA-PK) complex, which plays a central role in the non-homologous end joining (NHEJ) pathway for repairing DNA double-strand breaks (DSBs). DNA-PKcs binds to broken DNA ends alongside Ku70/Ku80 heterodimers, facilitating the recruitment of repair factors and stabilizing damaged DNA during repair. PRKDC is essential for maintaining genomic stability, V(D)J recombination in immune cells, and telomere maintenance.

Researchers use PRKDC antibodies in techniques like Western blotting, immunohistochemistry, and immunofluorescence to study its expression, localization, and interactions in cellular contexts. Dysregulation of PRKDC is linked to cancer progression, as its overexpression in tumors often correlates with resistance to radiation or chemotherapy. Conversely, mutations in PRKDC can result in severe combined immunodeficiency (SCID) due to defective lymphocyte development. PRKDC inhibitors are also explored in oncology to sensitize cancer cells to DNA-damaging therapies. The antibody thus serves as a vital tool for investigating DNA repair mechanisms, immune disorders, and therapeutic strategies targeting genomic instability.
