The GYS1 antibody is a crucial tool for studying glycogen synthase 1 (GYS1), a rate-limiting enzyme encoded by the *GYS1* gene that catalyzes glycogen synthesis by forming α-1.4-glycosidic linkages in glucose polymers. GYS1 is predominantly expressed in skeletal muscle and other tissues, while its isoform, GYS2. is liver-specific. Dysregulation of GYS1 is linked to metabolic disorders and cancer. Mutations in *GYS1* are associated with glycogen storage disease type 0 (GSD0), characterized by hypoglycemia and muscle weakness. In cancer, GYS1 overexpression has been observed in hepatocellular carcinoma, breast cancer, and gliomas, where aberrant glycogen metabolism supports tumor proliferation and survival under hypoxic or nutrient-deprived conditions.

GYS1 antibodies enable the detection and quantification of GYS1 protein levels in research applications such as Western blotting, immunohistochemistry (IHC), and immunofluorescence (IF). These antibodies help investigate GYS1's role in metabolic reprogramming, a hallmark of cancer, and its interplay with signaling pathways like AKT/mTOR. Commercial GYS1 antibodies are typically raised in rabbits or mice, with validation in specific experimental models. Recent studies also explore GYS1 inhibition as a therapeutic strategy to disrupt cancer cell energy metabolism. However, cross-reactivity with GYS2 or other isoforms requires careful validation. Overall, GYS1 antibodies are vital for advancing research in glycogen metabolism, metabolic diseases, and oncology.
