The STAT6 (Signal Transducer and Activator of Transcription 6) protein is a member of the STAT family of transcription factors, which play critical roles in mediating cellular responses to cytokines and growth factors. STAT6 is primarily activated by interleukin-4 (IL-4) and interleukin-13 (IL-13) through the JAK-STAT signaling pathway. Upon cytokine binding to receptors, STAT6 is phosphorylated, dimerizes, and translocates to the nucleus to regulate the expression of target genes involved in immune responses, cell proliferation, and differentiation. It is essential for T helper 2 (Th2) cell development, allergic inflammation, and alternative activation of macrophages.

STAT6 antibodies are widely used in research to detect and study STAT6 expression, activation, and function in various biological contexts. These antibodies enable the identification of STAT6 in techniques such as Western blotting, immunohistochemistry (IHC), immunofluorescence (IF), and flow cytometry. In clinical diagnostics, STAT6 antibodies are particularly valuable for diagnosing solitary fibrous tumors (SFTs), as nuclear expression of STAT6 (due to NAB2-STAT6 gene fusion) serves as a sensitive and specific marker. Dysregulation of STAT6 has also been implicated in cancers, autoimmune diseases, and allergic disorders, making it a therapeutic target. Researchers rely on validated STAT6 antibodies to explore its role in disease mechanisms and potential interventions. However, antibody specificity and batch consistency require careful validation to ensure accurate experimental results.
