The Annexin A2 receptor (ANXA2R), also known as AXIIR or p11. is a cell-surface protein that binds Annexin A2 (ANXA2), a calcium-dependent phospholipid-binding protein involved in membrane organization, vesicle trafficking, and extracellular proteolysis. ANXA2R facilitates the interaction of ANXA2 with tissue plasminogen activator (tPA) and plasminogen, playing a key role in fibrinolytic activity and cell surface proteolysis. This receptor is implicated in diverse physiological processes, including angiogenesis, cell migration, and immune response regulation.

In pathological contexts, ANXA2R is often overexpressed in cancers (e.g., breast, pancreatic, glioblastoma) and associated with tumor progression, metastasis, and therapeutic resistance. Its role in promoting plasmin generation contributes to extracellular matrix degradation, facilitating invasive behavior. ANXA2R antibodies are critical tools for studying these mechanisms, enabling detection of receptor expression in tissues or cell lines via techniques like immunohistochemistry, flow cytometry, or Western blot. Researchers also use blocking antibodies to inhibit ANXA2R-ANXA2 interactions, exploring their therapeutic potential in cancer and thrombotic disorders. However, ANXA2R's dual roles in homeostasis and disease necessitate context-specific evaluation. Recent studies highlight its involvement in viral entry (e.g., SARS-CoV-2) and autoimmune diseases, expanding its biomedical relevance. Validated antibodies with high specificity remain essential for clarifying its multifunctional biology and translational applications.
