S1PR5 (sphingosine-1-phosphate receptor 5) is a G protein-coupled receptor that binds sphingosine-1-phosphate (S1P), a bioactive lipid regulating immune cell trafficking, vascular function, and neural signaling. As the least characterized member of the S1PR family (S1PR1-5), S1PR5 is primarily expressed in natural killer (NK) cells, oligodendrocytes, and specific brain regions. It plays roles in immune modulation by influencing NK cell migration and retention in lymphoid organs, as well as in maintaining central nervous system (CNS) homeostasis, particularly in myelination processes.

S1PR5-targeting antibodies are research tools or potential therapeutics designed to modulate receptor activity. They can act as antagonists to block S1P-S1PR5 signaling or agonists to mimic its effects. Such antibodies are investigated in preclinical models of autoimmune diseases, cancer (via NK cell regulation), and neurodegenerative disorders like multiple sclerosis. Unlike S1PR1-targeting drugs (e.g., fingolimod), S1PR5-selective modulation may offer reduced cardiovascular side effects. However, challenges remain in achieving receptor specificity due to structural similarities among S1PR subtypes. Current studies also explore S1PR5 antibodies as biomarkers for neurological conditions. Their therapeutic potential is enhanced by emerging structural data from cryo-EM studies of S1PR5-ligand interactions.
