ROS1 (c-ros oncogene 1) antibodies are critical tools in cancer research and diagnostics, particularly in identifying ROS1 rearrangements associated with oncogenesis. Discovered in 1986. ROS1 encodes a receptor tyrosine kinase (RTK) involved in cell differentiation, proliferation, and survival. While normally silent in most adult tissues, chromosomal rearrangements (e.g., CD74-ROS1 fusion) lead to constitutive kinase activation, driving tumor growth in cancers like non-small cell lung cancer (NSCLC), glioblastoma, and cholangiocarcinoma.

ROS1 antibodies are designed to detect either the ROS1 protein or specific fusion variants. In clinical practice, immunohistochemistry (IHC) using ROS1 antibodies serves as a cost-effective screening method to identify patients eligible for targeted therapies like crizotinib or entrectinib. These antibodies must exhibit high specificity to minimize cross-reactivity with structurally similar kinases like ALK or MET. Companion diagnostic assays using validated ROS1 antibodies are essential for guiding precision oncology decisions.

Research-grade ROS1 antibodies also facilitate mechanistic studies of ROS1 signaling pathways and drug resistance mechanisms. Challenges remain in optimizing antibody sensitivity for low-abundance fusion proteins and standardizing detection protocols across laboratories. As ROS1-targeted therapies expand, the development of next-generation ROS1 antibodies continues to play a pivotal role in advancing both diagnostic accuracy and therapeutic strategies for ROS1-driven cancers.
