TNIP3 (TNFAIP3-interacting protein 3), also known as ABIN3. is a ubiquitin-binding protein that regulates inflammatory signaling pathways, particularly those mediated by NF-κB. It shares structural homology with other ABIN family members (TNIP1. TNIP2) and contains conserved domains, including a ubiquitin-binding domain (UBAN) and an NF-κB essential modulator (NEMO)-binding domain. TNIP3 interacts with TNFAIP3 (A20), a critical negative regulator of NF-κB, to modulate immune responses, apoptosis, and cell survival. Dysregulation of TNIP3 has been implicated in autoimmune diseases, cancers, and inflammatory disorders.

Antibodies targeting TNIP3 are essential tools for investigating its expression, localization, and molecular interactions. They enable researchers to study TNIP3's role in fine-tuning NF-κB activation, either by stabilizing A20's deubiquitinase activity or through competitive binding with other signaling components. Western blotting, immunohistochemistry, and co-immunoprecipitation are common applications. Studies using TNIP3 antibodies have revealed tissue-specific expression patterns and disease-associated alterations, such as reduced levels in lupus or aberrant expression in certain lymphomas. However, challenges remain in distinguishing TNIP3 from homologous proteins due to sequence conservation, necessitating rigorous antibody validation. These reagents continue to advance our understanding of TNIP3's dual roles as a tumor suppressor and context-dependent oncogenic collaborator.
