PAWR (Prostate Apoptosis Response-4), also known as Par-4. is a tumor suppressor protein encoded by the *PAWR* gene. Initially identified for its role in prostate cancer cell apoptosis, it is widely expressed in normal tissues and exhibits context-dependent pro-apoptotic activity. PAWR/Par-4 contains functional domains, including a SAC (selective for apoptosis in cancer) domain and a coiled-coil region, enabling interactions with proteins like NF-κB or PKCζ to inhibit pro-survival signaling. Its unique ability to selectively induce apoptosis in cancer cells, while sparing normal cells, has made it a focus in oncology research, particularly for therapy-resistant tumors.

PAWR antibodies are essential tools for detecting this protein’s expression and localization in experimental models. They are used in techniques such as Western blotting, immunohistochemistry, and immunofluorescence to study PAWR’s role in cancer progression, metastasis, and response to therapies. Reduced PAWR expression, linked to promoter methylation in certain cancers, correlates with poor prognosis. Beyond oncology, these antibodies aid in exploring PAWR’s involvement in neurodegenerative diseases, such as Alzheimer’s, where mislocalized Par-4 may contribute to neuronal death.
