The leukocyte-associated immunoglobulin-like receptor 2 (LAIR2) is a soluble protein belonging to the immunoglobulin superfamily, closely related to its membrane-bound counterpart, LAIR1. Both receptors interact with collagenous motifs, but LAIR2 lacks transmembrane and cytoplasmic domains, functioning instead as a decoy receptor. Discovered in the early 2000s, LAIR2 is primarily expressed in immune cells and placental tissues. It binds collagen with high affinity, competing with LAIR1 to modulate immune inhibitory signaling pathways. By blocking LAIR1-collagen interactions, LAIR2 indirectly enhances immune activation, contrasting LAIR1’s role in suppressing excessive immune responses.

Research highlights LAIR2’s involvement in immune regulation, particularly in contexts like cancer, autoimmune diseases, and infections. In tumors, LAIR2 overexpression may disrupt LAIR1-mediated immunosuppression, potentially promoting anti-tumor immunity. Conversely, elevated LAIR2 levels in autoimmune conditions could exacerbate inflammation by overriding immune checkpoint signals. LAIR2 antibodies, developed for research and therapeutic exploration, target its collagen-binding domain to study these mechanisms or manipulate immune responses. Recent studies also explore LAIR2 as a biomarker for disease progression or therapeutic response. Despite its emerging significance, LAIR2’s precise physiological and pathological roles remain under investigation, with ongoing efforts to harness its regulatory functions for immunotherapy strategies.
