The CBX3 antibody targets the Chromobox homolog 3 (CBX3) protein, a member of the Polycomb group (PcG) family involved in epigenetic regulation. CBX3. also known as HP1γ (Heterochromatin Protein 1 gamma), is one of three mammalian HP1 isoforms (α, β, γ) that play critical roles in chromatin organization, gene silencing, and maintenance of heterochromatin. It binds to trimethylated lysine 9 on histone H3 (H3K9me3), a hallmark of transcriptionally repressive chromatin, facilitating the formation of compact chromatin structures. CBX3 interacts with diverse proteins, including SUV39H1 methyltransferases and transcriptional co-repressors, to regulate processes like DNA repair, cell cycle progression, and telomere stability.

CBX3 antibodies are widely used in research to investigate its role in development, differentiation, and diseases such as cancer. In cancer biology, CBX3 exhibits dual roles, acting as either an oncogene or tumor suppressor depending on context, making it a focus for therapeutic exploration. These antibodies are essential tools in techniques like Western blotting, immunofluorescence, chromatin immunoprecipitation (ChIP), and flow cytometry to assess protein expression, localization, and chromatin interactions.

Commercial CBX3 antibodies are typically raised in rabbits or mice using immunogenic peptide sequences specific to the protein’s conserved chromodomain or C-terminal regions. Validation includes knockout cell lines or siRNA knockdown to confirm specificity.
