The cholecystokinin B receptor (CCKBR), also known as CCK-B receptor or CCK2R, is a G protein-coupled receptor (GPCR) primarily activated by gastrin and cholecystokinin (CCK). It plays a critical role in regulating gastric acid secretion, cell proliferation, and digestive processes. Structurally, CCKBR contains seven transmembrane domains and interacts with Gq proteins, triggering intracellular signaling pathways like phospholipase C (PLC) activation and calcium mobilization.

CCKBR is expressed in the stomach, brain, and certain cancers. Overexpression of CCKBR has been linked to gastrointestinal disorders (e.g., Zollinger-Ellison syndrome) and malignancies, including gastric, colorectal, and pancreatic cancers, where it promotes tumor growth and metastasis. In the central nervous system, it modulates anxiety, memory, and pain perception.
