名称	Vepdegestrant
描述	Vepdegestrant (ARV-471) is a selective and highly potent estrogen receptor (ER, ESR1) PROTAC degrader with strong degrading activity against the ER protein. By directly degrading the ER protein rather than merely antagonizing its activity, vepdegestrant effectively inhibits the ER signaling pathway and demonstrates significant antitumor activity in ER-positive (ER⁺) breast cancer, with a DC50 value of approximately 2 nM. It is particularly effective against tumors harboring ESR1 mutations.
体外活性	方法：MCF7 细胞加入 Vepdegestrant 100 nM，处理 0-24 小时，Western Blot 检测 ER 表达量。结果：ER 降解迅速，4 小时内 ER 降解 >80%。[1]
体内活性	方法：MCF7 乳腺癌细胞原位异种移植小鼠，Vepdegestrant: 10 mg/kg/d 口服，每日一次，连续 3 天，给药后 18 小时处死，取肿瘤组织做 Western Blot 检测 ER 水平。结果：Vepdegestrant 引起瘤内 ER 水平降低 ≥90%。[1] 方法：以 Vepdegestrant (ARV-471) 处理 MCF7、T47D 等 ER + 乳腺癌细胞，联合 10 ng/mL IFN-γ 刺激 24 h，流式细胞术检测细胞表面 MHC-I 表达。结果：Vepdegestrant 作为新型选择性雌激素受体降解剂（SERD），可显著逆转雌激素对 IFN-γ 诱导 MHC-I 表达的抑制作用，有效上调抗原呈递关键分子水平。[2]
