名称	GRI918013
描述	GRI918013 is a compound that selectively and competitively inhibits the autocrine motility factor (ATX/NPP2), thereby suppressing tumor cell invasion and metastasis. GRI918013 binds competitively to the active site of ATX, preventing the entry of lipid substrates such as lysophosphatidylcholine (LPC). This reduces the ATX-catalyzed hydrolysis of LPC to form lysophosphatidic acid (LPA), thereby inhibiting cell invasion and metastasis associated with the ATX-LPA axis. GRI918013 exhibits an inhibitory effect on the ATX-catalyzed hydrolysis of the LPL substrate FS-3, with an IC50 of 31.42 nM and a Ki of 12.98 nM. GRI918013 can serve as a tool for studying invasion and metastasis in cancers such as melanoma, and is also suitable for research on ATX-LPA axis-related diseases, including fibrotic diseases, neuropathic pain, and cholestatic pruritus.
体外活性	方法：将A2058人黑色素瘤细胞与GRI918013、ATX及LPC共同预孵育后，接种于Transwell上室，下室中加入趋化剂处理16小时，随后检测侵袭至下室的细胞荧光强度。结果：GRI918013可显著抑制A2058细胞侵袭，其IC₅₀值为118.79 ± 62.9 nM [1]。
体内活性	方法：为评估 GRI918013 的抗肿瘤转移作用，建立 B16-F10 小鼠黑色素瘤细胞实验性肺转移模型。取 C57BL/6 小鼠，经尾静脉注射 B16-F10 细胞，于肿瘤细胞接种后第1天开始腹腔注射给予 GRI918013 (30 μg/只)，每日1次，连续给药10天。于接种后第21天处死小鼠，解剖取肺组织，计数肺部表面转移结节数目。结果：GRI918013处理显著减少了肺转移结节数。[1]
