Bioactivity

名称	Natalizumab
描述	Natalizumab is a recombinant humanized monoclonal antibody and a humanized monoclonal antibody inhibitor that selectively targets α4 integrin (CD49d). Natalizumab binds to the α4β1 heterodimer and blocks its interaction with vascular cell adhesion molecule 1. Natalizumab also prevents lymphocytes from entering the central nervous system, thereby preventing acute demyelinating relapses, and is used in the study of relapsing-remitting multiple sclerosis and Crohn’s disease. Natalizumab possesses anti-inflammatory and immunomodulatory activity, inhibiting the adhesion, retention, and transendothelial migration of immune cells, and reducing the infiltration of inflammatory cells into the central nervous system or affected sites. Natalizumab is also being studied for autoimmune or inflammation-related diseases such as B-cell lymphoma and non-infectious uveitis.
体外活性	方法：健康供者（HD）的 PBMC，分选出 CD4+和 CD8+ T 细胞，加入 Natalizumab 2 μg/mL，预处理 30 分钟或未处理或同型对照 IgG4，细胞接种于 VCAM-1 包被板后刺激 10 分钟，高通量成像检测细胞黏附数、细胞面积、宽长比、pSLP76 强度、F-actin 强度。结果：Natalizumab 处理显著减少 T 细胞在 VCAM-1 上的黏附和铺展，降低 pSLP76 和 F-actin 强度。PCA 分析能清晰区分处理和未处理样本。[1] 方法：体外检测 Natalizumab 与淋巴细胞表面 VLA-4（α₄β₁整合素 / CD49d）的结合，体内观察其对白细胞跨血脑屏障迁移的影响。结果：Natalizumab 特异性结合 CD49d，阻断 α₄β₁与 VCAM-1 的相互作用，抑制淋巴细胞、B 细胞等向中枢神经系统（CNS）浸润。[2]
体内活性	方法：PTEN-CaP8 前列腺癌细胞荷瘤与 ADT 处理的 AppKI 小鼠给予 Natalizumab （62.5μg，腹腔注射，每周 2 次），同型 IgG 为对照。结果：Natalizumab 有效阻断 α4 整合素介导的免疫细胞黏附与迁移且 Natalizumab 修复 BBB 损伤、减少脑内免疫浸润、减轻神经炎症与胶质增生，显著改善 ADT 加重的认知缺陷。[3]
