Bioactivity

名称DL-Sulforaphane N-acetyl-L-cysteine
描述DL-Sulforaphane N-acetyl-L-cysteine (SFN-NAC), a metabolite of sulforaphane, induces apoptosis through down-regulation of α-microtubulin and phosphorylation of ERK1/2-mediated Stathmin-1, and up-regulation of Hsp70 through phosphorylation of ERK1/2 in non-small-cell lung cancer (NSCLC).
体外活性在对U87MG和U373MG细胞的研究中，使用DL-Sulforaphane N-acetyl-L-cysteine（30μM）处理24小时后，细胞周期分析显示药物诱导细胞在G2/M期停滞，并同时触发凋亡。在相同细胞系中，WesternBlot分析结果表明，DL-Sulforaphane N-acetyl-L-cysteine在10至50μM浓度范围内能够激活ERK1/2（Thr202/Tyr204），下调α-tubulin，并以剂量依赖的方式诱导自噬。此外，细胞活力实验结果显示，DL-Sulforaphane N-acetyl-L-cysteine在0至90μM浓度范围内，以剂量依赖的方式降低HA细胞、U87MG细胞和U373MG细胞的细胞活力，其IC50值分别为60.08μM、35.20μM和39.11μM[1]。
体内活性在HDAC抑制模型中，DL-Sulforaphane N-acetyl-L-cysteine（10 μmol，口服灌胃，单剂量，6小时处理）显著抑制了小鼠结肠黏膜中的组蛋白去乙酰化酶（HDAC）活性[2]。
