Bioactivity

名称Datopotamab deruxtecan
描述Datopotamab deruxtecan (DS-1062) is an antibody-drug conjugate (ADC) that targets tropomyosin 2 (TROP2) on the surface of trophoblast cells. Datopotamab deruxtecan exhibits antitumor activity by specifically binding to TROP2 and internalizing, releasing DXd to inhibit topoisomerase I (IC₅₀ = 0.31 μM), thereby inducing DNA damage and apoptosis. Datopotamab deruxtecan induces significant antibody-dependent cellular cytotoxicity in the presence of peripheral blood lymphocytes and inhibits tumor growth in xenograft models.
体外活性方法：在 USC 细胞系中，加入浓度梯度（0.01， 0.1， 0.5， 5.0， 50 μg/mL） Datopotamab deruxtecan，孵育 72 h，然后使用流式细胞术检测细胞活力。 结果：TROP2 高表达细胞对 Dato-DXd 高度敏感，低表达细胞敏感性差。[1]
体内活性方法：SCID 小鼠皮下接种 TROP2 高表达 USC 细胞。成瘤后分组：Datopotamab deruxtecan、对照 ADC、datopotamab 裸抗、溶剂对照。在第 0 天一次性眶后静脉丛给药(10 mg/kg)，饲养 50 天。 结果：与对照组相比，Datopotamab deruxtecan 治疗组小鼠的肿瘤生长被显著抑制，且总生存期显著延长。[1] 方法：Datopotamab deruxtecan 以 6 mg/kg 剂量静脉注射给雄性食蟹猴，在给药后 21 天内测量了血浆中 Datopotamab deruxtecan、总抗体和 deruxtecan 的浓度，以及肿瘤中的 deruxtecan 浓度。 结果：肿瘤中 deruxtecan 浓度高且持续，而血浆中浓度低，表明药物高效递送至肿瘤。[2]
