名称22-NBD Cholesterol
描述22-NBD Cholesterol (Fluoresterol) is a specific fluorescent probe that mimics cholesterol binding properties. It cannot be administered orally and is unable to cross the blood-brain barrier. 22-NBD Cholesterol (Fluoresterol ) specifically binds to cholesterol transport-related proteins such as ABCA1 and ABCG1, and is primarily used in cholesterol metabolism research, particularly for the visualization and quantitative analysis of cholesterol absorption, efflux, intracellular transport efficiency, and reverse cholesterol transport (RCT). The excitation and emission wavelengths of 22-NBD Cholesterol are 472 nm and 540 nm, respectively. Furthermore, 22-NBD Cholesterol can be used to investigate cholesterol metabolism mechanisms in diseases such as hyperlipidemia, atherosclerosis, and non-alcoholic fatty liver disease (NAFLD).
体内活性方法：将负载 22-NBD Cholesterol 胆固醇的 RAW264.7 巨噬细胞经腹腔注射入 P407 诱导的高脂血症小鼠体内，24 h 后收集血液、肝脏、胆汁及粪便，检测荧光强度以评估胆固醇逆向转运（RCT）。 结果：与对照组相比，P407 促进标记胆固醇在肝脏中聚集。EEDL 处理显著降低小鼠肝脏和血清中的 22-NBD Cholesterol 水平，而对胆汁中的含量无显著影响。此外，P407 降低粪便荧光强度，而 EEDL 显著增加粪便中胆汁酸分泌，表明 EEDL 可促进巨噬细胞依赖的 RCT 过程[1]。
