名称	Z-IETD-FMK
描述	Z-IETD-FMK (Z-IE(OMe)TD(OMe)-FMK) is a cell-permeable, irreversible inhibitor specific for caspase-8. Z-IETD-FMK also an inhibitor of granzyme B. Z-IETD-FMK exhibits cell permeability. Z-IETD-FMK can be used in studies of the extrinsic apoptosis pathway, pyroptosis, and programmed necrosis.
激酶实验	Proteasome assay: Exponentially growing cells on a 96-well clustered plate are treated with different concentrations of drugs or left untreated (control) for 6 hours. Proteasomes extracted with 0.5% NP40 buffer are mixed with equal amounts of samples in 100 μL total volume, and then incubated with 25 μmol/L of fluorogenic substrates (LRR- specific for trypsin-like activity, LLE-specific for caspase-like activity, and SUVY-specific for chymotrypsin-like activity) in black-bottom 96-well plates at 37°C. Fluorescence is monitored every 5 minutes at the wavelength of 360 nm (excitation) and 480 nm (emission).
体外活性	方法：人 PBMCs、纯化的 CD4⁺和 CD8⁺ T 细胞加入丝裂原（PHA 5 μg/mL）和 Z-IETD-FMK （25， 50， 100 μM），处理 72 小时， [³H]-胸腺嘧啶掺入法检测 DNA 合成；CFSE 稀释法流式细胞术检测细胞分裂。结果：Z-IETD-FMK 在 100 μM 时显著抑制 T 细胞增殖。[1] 方法：小鼠骨髓来源中性粒细胞，加入 50 μM Z-IETD-FMK，24 小时（ELISA）；6-8 小时（qPCR），ELISA 检测培养上清中 CXCL1 和 IL-1β水平，qRT-PCR 检测 IL-1β和 TNF-α mRNA。结果：Z-IETD-FMK 显著诱导中性粒细胞产生 CXCL1 和 IL-1β，并上调其 mRNA。[2]
体内活性	方法： C57BL/6 小鼠，经鼻感染肺炎链球菌 D39 或耐碳青霉烯肺炎克雷伯菌，感染后 24 小时开始静脉注射 Z-IETD-FMK (6 mg/kg)，每日一次，持续 5 天。结果：Z-IETD-FMK 治疗显著提高生存率(肺炎链球菌：对照组 38% vs 治疗组 88%)，显著降低肺内细菌载量。[2] 方法：成年雄性 SD 大鼠，血管内穿孔法建立 SAH 模型。从 SAH 后 21 天开始尾静脉注射 (Z-IETD-FMK) 1 mg/kg，持续至 27 天。结果：Z-IETD-FMK 治疗组在空间学习和记忆能力受损，运动协调性下降均显著改善，表明其对长期神经功能有保护作用。[3]
