名称	Canagliflozin hemihydrate
描述	Canagliflozin hemihydrate (TA 7284) is a drug of the gliflozin class or subtype 2 sodium-glucose transport inhibitors used for the treatment of type 2 diabetes. SGLT2 is responsible for at least 90% of renal glucose reabsorption (SGLT1 being responsible for the remaining 10%). Blocking this transporter causes up to 119 grams of blood glucose per day to be eliminated through the urine, corresponding to 476 kilocalories.
体外活性	Canagliflozin 在 CHO-hSGLT2 细胞中抑制与 Na+ 依赖的 14C-AMG 吸收，其 IC50 为 4.4±1.2 nM。在 CHO-rSGLT2 及 CHO-mSGLT2 细胞中获得了相似的 IC50 值（分别为大鼠和小鼠 SGLT2 的 IC50 = 3.7 和 2.0 nM）。Canagliflozin 还在 CHO-hSGLT1 和 mSGLT1 细胞中抑制 14C-AMG 吸收，IC50 分别为 684±159 nM 和 >1,000 nM。
体内活性	Canagliflozin（30 mg/kg，为期4周的治疗）在DIO小鼠中降低了血糖（BG）水平、呼吸交换比率和体重增加[1]。与接受载体处理的大鼠相比，Canagliflozin（3 mg/kg，为期3周）增加了尿糖排泄（UGE），而总食物摄入量没有显著变化，导致ZF大鼠体重下降[1]。
