Bioactivity

名称	Busulfan
描述	Busulfan is an alkylating antineoplastic agent derived from dimethane sulfonate, with cytotoxic and immunosuppressive properties. It primarily acts by forming carbonium ions in vivo, which induce cross-linking between DNA strands or between DNA and proteins, leading to DNA damage, inhibition of DNA replication, and suppression of RNA transcription. In addition, Busulfan can inhibit thioredoxin reductase and induce apoptosis. It is commonly used as a myeloablative agent in preconditioning regimens for bone marrow transplantation and can also be used to establish anemia models.

体内活性	Busulfan,一种通过交联DNA和DNA和蛋白引起DNA损伤的烷化剂,通过细胞外信号调节激酶（Erk）和p38丝裂原活化蛋白激酶（p38 MAPK）诱导正常人二倍体WI38成纤维细胞衰老级联独立于p53-DNA损伤途径。Busulfan诱导谷胱甘肽的短暂减少,但ROS产量会持续增加。Busulfan诱导的Rb低磷酸化通过抑制睾丸细胞中的PCNA表达来防止精原干细胞的凋亡。Busulfan抑制鹅卵石区域形成细胞的频率,但不会引起造血干细胞相似的细胞和祖细胞的细胞凋亡明显增加。Busulfan通过依赖细胞凋亡的机制抑制HSC类似细胞和祖细胞的造血功能。Busulfan诱导骨髓造血细胞衰老与p16Ink4a和p19Arf表达增加有关,这具有时间依赖性。
